Realtime visualizing brain cells with a fluorescent chemical probe
The brain is a mysterious organ of our body. Recent knowledge has led us to think about glia as well as neurons to understand the brain. However, the lack of easy methods for tracking a type of glia and/or neurons makes a requirement of vast effort for developing genetically modulated animals. Herein, I will present the development of a chemical fluorescent probe for microglia, which enables the visualizing and tracking of microglia at tissue levels by adding the compound just before observation. The developed probe, named CDr20, even can detect microglia in vivo through simple intravenous injection if BBB has slightly altered. The mechanism study revealed that CDr20 label microglia through the Ugt1a7c-mediated fluorescence turn-on process in microglia. Because of its simplicity and specificity, developing novel fluorescent chemical probes for targeting other types of brain cells by the described platform will be very useful in basic neuroscience and further in biomedical applications.
